Plant Regeneration through Somatic Embryogenesis in Zoysiagrasses (Zoysia spp .)



Abstract
Experiments were carried out to develop methods for the regeneration of plants through somatic embryogenesis in zoysiagrasses. Immmature inflorescences of various lengths (¡Â 5, 6-10, 11-15, 16-20, ¡Ã20 §®) and caryopses at different maturities (1, 2, 3, 4 weeks after pollination) were cultured on MS or N6 agar medium supplemented with 2 §·/L of 2,4-D. The most embryogenic calli were induced from 3-week-old immature caryopses cultured on media containing 2 §·/L of 2,4-D. The 2,4-D requirement could be lowered to 1 §·/L by adding 20 g/L of sorbitol to produce and maintain the embryogenic calli. When the calli were cultured in the liquid MS medium with agitation, some proliferated rapidly as compact cell clumps of proembryo body. Numerous plantlets regenerated simultaneously upon transfer to auxin-free MS medium. The culture method was also effective in other genotypes like Z. sinica and an interspecific hybrid between Z. sinica and Z. matrella.
